Principle
GenSampler, is an automated system that enables a sequential sampling of drug-producing cells directly from the bioreactor without shear stress. Cells are then mixed with lysis buffer, which allows to extract and analyze the RNA in a separate experiment to identify the biomarkers of interest.
Description
GenSampler combines novel and outstanding technologies that are interconnected in order to ensure a safe sampling of cells.
Priming solution: used to clean the tubing before & after using + between each sampling
Rinsing solution: used to rinse and clean the tubes after each sampling
Detergent solution: concentrated detergent used to clean the tubing after each sampling and after usage
Trash: used to get rid of residual and dead volumes.
Trapping system: encompasses many elements. The bioreactor is connected in a sterile way to the GenSampler via an aseptic connection. Cells are trapped via an acoustic trap where they are filmed and automatically counted . When the desired number of cells is reached, the sample is introduced into a septum tube via a specific needle. Twelve cell samplings can be made and are separated by a washing and lysis steps
Lysis system: The trapping process is followed by a lysis step where cells are mixed with lysis buffer.
Independently and following this step, RNAs are extracted and analyzed to check the expression level of specific genes. This service is provided as well by GenSensor using a biomarker pipeline that has been developed by our team.
Electrical system:
a complexe electrical part integrates and covers all the mechanical and informatic parts of the GenSampler and enables a robust operation of the machine.
